The Queen's Street area of Enniskillen was re-opened to traffic later on Saturday following the serious road accident in the early hours of the morning.
Police say that a blue Ford Focus believed to be carrying six people, was involved in a collision with the Johnston Bridge Roundabout after failing to stop for police a short time beforehand.
A 23-year-old man was taken to hospital with serious head injuries and two other men, aged 28 and 27 and two women aged 42 and 22 were taken to hospital for treatment to injuries not thought to be life threatening.
An 18-year-old man was arrested and is currently in custody assisting police with enquiries.
Inspector Rod Flood from Enniskillen PSNI Station has appealed for anyone who witnessed the incident or anyone who was travelling on the Cornagrade Road around the time of the incident to contact them on the 101 number.
