The Killadeas Road between Trory Roundabout and Killadeas is currently closed as emergency services deal with a road traffic collision involving two vehicles.
The PSNI are advising road users to divert through Irvinestown until further notice.
A PSNI spokeswoman said: "Police are currently in attendance at a road traffic collision involving two vehicles at Killadeas Road in Enniskillen.
"Diversions are in place and motorists are advised to seek alternative routes."
